It’s Certain Someone! Talk about giving your girl some lip this week. Well, well, I’ll hold my tongue and pretend I didn’t get the sarcasm/spite behind your words. lol. Dirt off this chick’s shoulder, certain someone. You’ll have to try harder. I’m learning to be happy with my lot for now. When the time comes, I’ll make my move.
Sorry for being cryptic, y’all. This is the first hit if you google for me so I’m not trying to air dirty laundry right now. In other news, I got word from an awesome relative of mine, EA. I’ve been MIA sadly. I hope that we can meet up some day. It’s been too long. Life at the lab’s going well. I had a mini-scare with a protein I was working with. Here’s what happened: this protein goes through two columns (an ion exchange column first and a gel filtration column next). After pooling the fractions containing my protein, it gets concentrated on the basis of size i.e. molecules above a certain molecular weight are retained. Then, we dialyze to get the protein in our buffer/solution of choice before injecting this protein into an HPLC machine (high pressure liquid chromatography). After this step comes determining the concentration of protein I’ve ‘purified’ and this is where my legs nearly turned to jello. The test for determining protein concentration is called a BCA assay and it’s a colorimetric assay. This means if there’s protein, it changes color. Well, I did the test once and there was no color change. I did it again and again. No luck. Well, this assay’s pretty sensitive (I’m not sure of the exact numbers), but we eventually had to do some more concentrating of my protein (from 5mls to about 500 microliters). I’ll repeat the BCA assay and hopefully get a more concentration protein solution reading. Wish me luck!
